Ancient-pathogen genomics as a window to the history and evolution of infectious diseases

Infectious diseases continue to be an important health problem at a worldwide scale, despite the existence of antibiotic treatments, vaccination, sanitary consciousness, etc. The advent of High-Throughput Sequencing (HTS) a decade ago, made possible not only to sequence full genomes from pathogens but also to access to the genomic diversity among their existing strains. This information has revolutionized the way we can study the biology of pathogens, how they change and evolve and to better understand their epidemiology. However, the information we can retrieve from modern pathogen genomes sometimes is not sufficient to determine how and when a pathogen emerged, how it spread and which genomic changes were more determinant for its virulence and pathogenicity. In the past few years, the study of ancient pathogen genomes using ancient DNA (aDNA) and HTS has demonstrated to provide key pieces of information towards these fundamental questions. In this talk, I will show how ancient pathogen genomics can be used to better understand the history and evolution of infectious diseases. I will use plague (Yersinia pestis infection) as a paradigmatic example, since this is the best studied case of ancient infectious diseases. I will make an overview of the lessons learned from the genomes of Y. pestis strains that caused multiple massive pandemics over the history of humanity. I will finally show how our recent discovery of the most ancient case of Y. pestis infection (5,000 years ago, in Sweden) made possible to build the best model to date of how this pathogen emerged and rapidly spread during the Neolithic and Bronze Age across Eurasia, leading to what probably was the first human pandemic of history.
