SHAPE: Saut d’Hôtes, Adaptation des Pathogènes et Emergence

            Malgré les avancées technologiques de la recherche scientifique et médicale au cours du 20ème siècle, les maladies infectieuses représentent la troisième cause de mortalité dans le monde (Organisation Mondiale de la Santé OMS).  Les maladies infectieuses constituent donc un fardeau considérable pour les populations humaines et animales. Parmi les maladies infectieuses touchant les populations humaines, 2/3 sont zoonotiques, c’est-à-dire issues d’un transfert d’un pathogène à partir de la faune sauvage ou domestique. De nombreuses questions entourent ce phénomène de saut d’hôtes multi-spécifiques, notamment comment et pourquoi certains pathogènes sont capables d’infecter une nouvelle espèce et de s’y maintenir ?

La transmission d’un pathogène entre espèces se réalise ainsi à travers la succession de plusieurs étapes conditionnées par des nombreux facteurs aussi bien de type sociologique, économique, écologique qu’évolutif. Trois étapes sont toutefois cruciales :

 - La première étape se situe au niveau du contact et de la rencontre entre l’agent pathogène et l’hôte susceptible.

 - Une fois l’hôte infecté, le pathogène va devoir s’adapter au système immunitaire de l’hôte pour survivre et se reproduire.

 - La troisième étape repose sur la diffusion du pathogène à une échelle populationnelle et est fonction du contact entre hôtes.

C’est au travers de ces différentes étapes que les travaux de recherche développés dans notre équipe tentent de mieux comprendre comment les pathogènes « sautent » d’un hôte à un autre et s’y adaptent. Le but étant d’aboutir à une meilleure compréhension des modalités de transferts et in fine de l’émergence et/ou de la réémergence de ces maladies infectieuses.

 

Organisation du groupe :


            Nos travaux de recherche s’articulent autour de modèles d’études variés tels que les parasites du genre Plasmodium, les virus de la fièvre aphteuse, le MERS CoV, etc., et d’approches intégratives et complémentaires. Plus particulièrement, de nombreux projets de recherche sont actuellement développés pour :

1/ Élucider comment les pathogènes du genre Plasmodium s’adaptent génétiquement à de nouveaux environnements hôtes (hommes, gorilles et chimpanzés), à travers une approche intégrée alliant génétique des populations, génomique comparative et fonctionnelle. Franck Prugnolle et Virginie Rougeron

 2/ Explorer les interactions hôte-pathogènes dans le but de définir la barrière d’espèce au cours de la transmission d’agents infectieux, par l’étude des interactions entre les ligands des parasites Plasmodium et les récepteurs des hôtes (homme, gorille, chimpanzés).  3/ Reconstruire l’histoire démographique de Plasmodium vivax et de Plasmodium falciparum, grâce aux outils de la génétique des populations et de la génomique. Virginie Rougeron et Franck Prugnolle

 4/ Suivre la dynamique épidémiologique des pathogènes circulant dans la faune sauvage grâce aux mouches hématophages, utilisées comme ‘seringues volantes’, grâce aux outils de screening moléculaire et d’analyse épidémiologique. Franck Prugnolle

 5/ Evaluation du potentiel zoonotique de Plasmodium vivax et Plasmodium vivax-like. Virginie Rougeron

 6/ Estimer les taux de contacts entre espèces et comprendre les mécanismes sociologiques et écologiques de ces interactions. Eve Miguel

 7/ Décrire la diffusion des pathogènes au sein des populations dans le temps et dans l’espace. Benjamin Roche et Eve Miguel

 8/Décrire les communautés d’hôtes pour définir la notion de réservoir et comprendre l’impact de la diversité d’hôtes sur la transmission des agents pathogènes, au travers d’études empiriques combinées avec des approches de modélisation mathématique. Benjamin Roche et Eve Miguel

 9/ Comprendre comment se structure les communautés d‘agents pathogènes et quelles sont les conséquences pour les stratégies de santé publique verticale, grâce au développement de méthodes statistiques sophistiqués combinés avec de la modélisation mathématique. Benjamin Roche

 10/ Développer des approches permettant l’optimisation des stratégies de santé publique contre les maladies endémiques dans les pays aux ressources économiques limitées ou les maladies émergentes dans les zones à forts revenues, grâce à l’intégration au sein de modèle mathématiques des connaissances en écologie, génétique, sociologie et épidémiologie. En plus de ces travaux de recherche, tous les membres de l’équipe sont amenés à être impliqués dans la formation des étudiants et postdoctorants, mais également dans l’enseignement dans des modules de Master à l’université de Montpellier mais également à l’international (Computational Biology for Infectious Diseases.
